Imaging Bile Canaliculi

چکیده
Analyzing transport of biliary metabolites is essential to predict pharmacokinetics and hepatotoxicity during drug development. A functional impairment of hepatobilary transporters, such as bile salt export pump (BSEP) and multidrug resistance-associated protein 2 (MRP-2), is strongly associated with an increased risk of cholestatic liver injury. Currently, artificial models, such as BSEP expressing membrane vesicles, are used for studying efflux transporter function. Sandwich-cultured hepatocytes also are utilized as an in vitro tool to study hepatobiliary drug transport and hepatotoxicity [Lepist et al., 2014]. However, these models lack the functional complexity of the natural 3-dimensional (3D) liver environment. 3D liver cells immediately display key morphological and functional characteristics of native hepatocytes, such as cuboidal cell shape, increased formation of bile canaliculi and higher drug efflux activity, compared to the conventional monolayer culture [Hammad et al., 2014; Mueller et al., 2011]. Phalloidin-induced alterations of bile canaliculi.
Phalloidin, a toxin from the plant Amanita phalloides, irreversibly polymerizes actin filaments and causes cholestasis. Three-dimensional structural changes induced by phalloidin in the bile canaliculi and the intra-acinar localization of these changes were studied in the rat liver by scanning and transmission electron microscopy. After 3 days of treatment, canalicular changes appeared mainly i...
متن کاملCurcumin Prevents Bile Canalicular Alterations in the Liver of Hamsters Infected with Opisthorchis viverrini
Opisthorchis viverrini infection causes inflammation and liver injury leading to periductal fibrosis. Little is known about the pathological alterations in bile canaliculi in opisthorchiasis. This study aimed to investigate bile canalicular alterations in O. viverrini-infected hamsters and to examine the chemopreventive effects of curcumin on such changes. Hamsters were infected with O. viverri...
متن کاملHow transfer rates generate Gd-BOPTA concentrations in rat liver compartments: implications for clinical liver imaging with hepatobiliary contrast agents.
Following the injection of hepatobiliary contrast agents, MRI detects all molecules included in a region of interest but cannot estimate true concentrations in sinusoids, interstitium, hepatocytes or bile canaliculi. The aim of the study was to measure true concentrations in hepatocytes and to show how transfer rates across sinusoidal and canalicular membranes generate these concentrations. We ...
متن کاملIn vivo dynamic metabolic imaging of obstructive cholestasis in mice.
We tried to image obstructive cholestasis by using a newly developed imaging system to measure the alterations of hepatobiliary function in living mice with their bile ducts ligated. A hepatic imaging window was installed on the upper abdomen soon after the mice underwent ligation of the common bile duct. On the next day, the mice received intravenous injection of rhodamine B isothiocyanate-dex...

Cholestasis may be defined as stagnation of bile within the intrahepatic or extrahepatic biliary tree. Biochemically, there is retention of conjugated bilirubin, bile salts, and alkaline phosphatase; morphologically, there is at least dilatation of biliary canaliculi and loss of their microvilli, and later accumulation of bile pigment in the canaliculi (bile plugs)."0 The mechanism underlying t...
